No study shows this formula turns back biological age. All four ingredients are at or above studied doses. Folic acid with vitamin B12 cut chromosome damage by about 25 percent over six months in young adults with high starting damage [1]. A high-dose active folate form lowered oxidative stress in older women [2], and folate improved antioxidant capacity and inflammation in women over 12 weeks [3]. A mix with folic acid and B12 lowered inflammation in older adults, though the effect cannot be credited to one vitamin [4]. Betaine at 3 grams raised one energy enzyme and one antioxidant enzyme but left other markers unchanged [5]. Against this, the largest trial, in 2,919 older adults, found B12 and folic acid did not slow physical decline or prevent falls [6]. Several trials found no change in the chemical tags on genes [7][8][9], and an aging clock fell only in a small subgroup of women with one gene variant [10]. Vitamin B6 lowered homocysteine but did not change oxidative stress or damage markers [11][12]. Animal and cell studies are never counted.

Customers across iHerb, Walmart, Vitacost and Woot, in the US, Europe, Australia and the Middle East, mostly say they trust the brand's quality. Many say they buy Thorne because of its reputation for clean, pure, gluten-free formulas, and a number of long-time buyers say they have used the brand for decades. A large group of reviewers have an MTHFR gene variant. They say the methylated forms suit them better than ordinary B vitamins, and some describe feeling calm and balanced. Reviewers often report more energy, better focus, mental clarity and mood. Others say they absorb B vitamins more easily. Several say they feel steadier or sharper in daily life, and some say they felt unwell when they ran out. A second theme is lab results. Many people were told to take the product by a doctor, nutritionist or specialist because of high homocysteine. They report that levels fell, in one case from around 11 to about 6. Some link this to concerns about family history or heart and brain health. A few with Hashimoto's disease or Hughes syndrome say it helps them keep symptoms to a minimum or improves their blood work. One reviewer in her 60s reported shedding water weight and gaining energy after starting. Reviewers do raise some complaints. The most common is price. Many call it expensive but say you get what you pay for. Some mention that the capsules are large. A few warn that methylated folate can cause a rough adjustment period, with several uncomfortable days. They advise starting slowly and continuing through it. One person with a B12 deficiency said the B12 dose was too low for them and they needed extra. A smaller group raises concerns about authenticity and ownership. One buyer was suspicious of bottles that looked different and was told by Thorne that only certain Amazon sellers are authorized. Another reviewer said they would stop buying because Procter & Gamble now owns Thorne. Overall, the feedback is strongly positive, and the criticism concerns cost, capsule size, sensitivity at the start, and trust in the company and its resellers.
Biohacker Summary

In this product every listed ingredient is a smaller ingredient, and folate drew the most comment, with all ten panelists touching on it in some way. Most of the panel recommends folate, and they prefer the methylated form (methylfolate) over synthetic folic acid, mainly to lower homocysteine and support the brain, mitochondria and pregnancy; one panelist takes the lower of two strengths once a day. Two of them warn that too much methylfolate can overstimulate or push levels above the normal range, and one noted that birth control pills and a med can deplete folate. Vitamin B12 got backing from several for mood, nerves and brain function, with methylcobalamin the favored form, though one warns against taking it too close to bedtime. Betaine and vitamin B6 were each mentioned by a few, with one panelist preferring the active form of B6 over a synthetic one.

- Folate is a methyl donor that can be obtained from leafy green vegetables and is important for DNA methylation.
- Recommends getting trimethylglycine (TMG), also known as betaine, from whole foods like beetroot, shrimps, spinach, wheat germ, wheat bran, mushrooms, and various vegetables to support DNA methylation.
Andrew Huberman · Vitamin B12 · neutral
- States that a deficiency in vitamin B12 can be terrible and lead to chronic fatigue-like syndromes, as many parts of mitochondria require B vitamins.
- States that B12, folate, and iron are essential to mitochondrial function and if deficient, mitochondria will not function properly, which is consistent with the mitochondrial theory of neuropsychiatric disorders.
- Recommends taking methyl B12 as part of methylated B vitamins to lower homocysteine levels and reduce cardiovascular risk.
- Recommends taking methyl folate as part of methylated B vitamins to lower homocysteine levels and reduce cardiovascular risk.
- Has been taking AG1 for over 10 years, once and often twice every day, to ensure he gets all necessary vitamins, minerals, probiotics, and adaptogens, considering it a nutritional insurance policy.
- Takes methyl folate because elevated levels of homocysteine are associated with negative outcomes, and taking methyl folate is known to lower homocysteine. This might lower ADMA and SDMA and raise nitric oxide synthesis, potentially impacting vascular disease.
- Does not supplement B6 due to evidence that overs-supplementing vitamin B, especially B6, is problematic because of peripheral nerve damage.
- Has been taking AG1 since 2012 to ensure he gets enough vitamins, minerals, Prebiotic fiber, and other nutrients typically found in fruits and vegetables, providing nutritional insurance.
- Andrew Huberman started drinking AG1 in 2012 and still drinks it once or twice a day because it provides all his foundational nutritional needs, ensuring proper amounts of vitamins, minerals, probiotics, and fiber for optimal mental health, physical health, and performance.
- Notes that some people are exploring the use of P5P, a metabolite of vitamin B6 known to suppress prolactin, as a way to buffer the post-MDMA crash, though there are no human data yet.
- Discusses betaine as a potential alternative for creatine non-responders, taken at 1 to 3 grams per day, which helps with amino acid synthesis and energy shunting.
- Mentions that there is some evidence that betaine, also called trimethylglycine, can improve lean mass and power output.
- Recommends the consumption of B12 and other B vitamins, especially folate, as it has been shown to partially decrease cancer risk in people who ingest alcohol, though it does not completely offset it.

RAL Summary
Members OnlyTier 4 for Healthy Aging
Tier 2 rests on vitamin B12 improving aging markers, but the B12 findings are thin and mixed: the largest trial found no benefit, gains came only from mixed formulas, and gene-tag and aging-clock results were mostly unchanged, so the studies disagree. So it is graded Tier 4, lower than the rules alone gave.
- Emerging: not enough evidence to rank higher yet.
Where the evidence disagrees: 18 of the 61 human studies found no effect.

Body systems: Cardiovascular system, Nervous system, Immune system
Combine with: For methylation and cellular health, Methyl-Guard pairs well with trimethylglycine (TMG) or choline for additional methyl donors, vitamin B6 as P5P and riboflavin (B2) to support homocysteine clearance, omega-3 fatty acids (EPA/DHA) for cardiovascular and cellular membrane support, and magnesium glycinate. Antioxidants such as vitamin C, vitamin E, and NAC or glutathione can complement it for oxidative stress. For fast heartburn relief, use a separate product such as calcium carbonate antacids, an alginate (sodium alginate) formula, or DGL licorice. Note that long-term acid-suppressing drugs (meds, H2 blockers) can lower B12 absorption, so methylcobalamin can be helpful for people using them.
Possible side effects: Some reviewers report a rough adjustment period of several uncomfortable days when starting methylated folate, and some find the capsules large. Betaine with other methylation supplements appeared safe in children over one year.
