Thorne Riboflavin 5'-Phosphate Review
Thorne Riboflavin 5'-Phosphate is a supplement that provides riboflavin in its active, tissue-ready form, R5P. The page describes riboflavin, also known as vitamin B2, as a nutrient that plays an integral role in activating other B vitamins. Because the body must first convert riboflavin to its bioactive form, riboflavin 5'-phosphate is presented as a more readily available option for use throughout the body than inactive riboflavin supplements. Thorne positions this product as a way to support the activation and conversion of other B vitamins, including B3 (niacin), B6, and folate. The listed benefits include helping to activate vitamin B6 and folate (B9), being necessary for the conversion of tryptophan to niacin (B3), supporting the metabolism of fats and glucose, helping to synthesize red blood cells, benefiting healthy methylation, and supporting the production and regulation of certain hormones. Each serving is one capsule containing 36.5 mg of riboflavin as riboflavin 5'-phosphate sodium, and the bottle contains 60 capsules. The other ingredients are microcrystalline cellulose, silicon dioxide, calcium laurate, and hypromellose capsule. The product is described as not derived from yeast, and it is presented as an easy-to-take protocol of one capsule per serving. The page says it may be suitable for people with compromised digestive systems, people who may be sensitive to yeast, people needing help with methylation processes, and people who may have issues activating vitamin B6 and or folate. It notes that a compromised digestive system can adversely affect the body's ability to convert riboflavin to R5P, and that certain meds can also compromise absorption of riboflavin, putting the body in a state of deficiency. Thorne states the formula reflects its scientific rigor, with each ingredient selected, clinically studied, and tested through multiple rounds of quality control for purity, potency, and consistency. The page notes it is trusted by health-care practitioners. It also states the product has been third-party tested and certified to verify that what is in the package matches the label and that it contains no unsafe levels of contaminants such as heavy metals, pesticides, and microorganisms. Warnings on the page state the product is contraindicated in individuals with a history of hypersensitivity to any of its ingredients, and that anyone who is pregnant should consult a health-care practitioner before using it. The page reports no known adverse interactions or contraindications at publication date.

This product gives riboflavin, vitamin B2, at 36.5 milligrams per capsule and 109.5 milligrams a day, far above the 1.3 milligrams that worked in a study. In a controlled trial lasting four weeks, construction workers who drank riboflavin-fortified apple juice every day reported less fatigue and had better antioxidant markers [1]. In adults with a rare disorder of fat metabolism linked to the drug a med, 38 of 39 treated with riboflavin improved, including less fatigue and fewer muscle symptoms [2]. In 10 patients with a late-onset form of this disorder, 400 milligrams daily quickly restored muscle strength, though nerve symptoms did not improve [3]. In 25 such patients, those who responded to riboflavin did well [4]. These disorder studies may not apply to healthy people. Two trials found no benefit, but both tested mixes of many nutrients rather than riboflavin alone: a nutrient mix did not reduce fatigue in 202 dialysis patients over 12 weeks [5], and a three-week antioxidant capsule did not preserve exercise performance at low oxygen in 17 people [6]. Animal and cell studies are never counted.

Across the stores that sell it, Thorne Riboflavin 5'-Phosphate carries about 3,053 ratings, and the feedback is strongly positive. Buyers mostly praise the quality, the small capsules and the active B2 form. A few found no effect or disliked side effects. **What buyers say it did for them:** Many report a real benefit. Some cite less fatigue, and others mention fewer headaches or migraines. One buyer wrote, "A couple days without it and I feel 10 times the stress." Another takes it three times a day on a neurologist's advice and says it "significantly helps some of my fatigue and fibromyalgia symptoms." One reviewer had pressure headaches stop after about a week: "have not had a pressure headache since!" Others mention gum health, red blood cell support, borderline-low blood work, and a B2 deficiency. Some buyers say its active form suits people who can't use regular B2 well. One reviewer said they could feel a difference within hours. **What they complain about:** Complaints are few. One buyer saw no change for headaches: "cannot see a difference yet for this purpose." The same person disliked that it turns urine bright yellow with an odor, which others also call normal but unpleasant. One reviewer got a med effect and suspects it would work better sublingually, and moved to another sublingual B complex. One buyer said the bottle is "not quite half full." One reviewer noted the product contains a few fillers, though of good quality. **Taste, smell, pill size and form:** The small capsule is the most repeated praise. One buyer wrote, "The capsules are very small and easy to swallow." Another called the pills "pretty small so they are easy to take with others." Another noted the capsules are very easy to open and mix with food. A reviewer on one store found the hypromellose capsule fine, and another said the taste is not great but acceptable. Another takes it on an empty stomach without trouble. **Dose:** Opinions on the dose are mixed. One reviewer found it high and takes only part of a capsule. Another splits it and says it works well that way. Another said, "Wish it came in smaller dose." Others call the dose good or not too high over the daily value. A few say it needs to be taken separately from other B vitamins. **Value for money:** Little is said about price. One buyer called it "Best overall in price and quality." Another said they stay with Thorne rather than a cheaper brand. Several say the bottle lasts a long time. **Who it seems to suit and who it did not:** It seems to suit people who trust Thorne and want a high-quality, bioavailable B2. That includes people with sensitive guts, those working with a neurologist, and those taking B12 injections, with whom one reviewer said it pairs perfectly. It suits people who want small capsules that are easy to swallow or open. It suited those looking for help with migraines or headaches, though not everyone saw that benefit. It suited people less if they are bothered by urine changes, want a lower dose, or prefer sublingual forms. Reviews of other forms of this product were left out.

RAL Summary
Members OnlyTier 4 for Energy
The ineffective grade rests on two trials of multi-nutrient or antioxidant mixes rather than riboflavin alone, while a controlled riboflavin trial reduced fatigue and reviews are strongly positive, which supports tier 3 rather than tier 5. So it is graded Tier 4, as the studies disagree.
- Emerging: not enough evidence to rank higher yet.
Where the evidence disagrees: 3 of the 7 human studies found no effect.

Body systems: Metabolic system, Muscular system, Nervous system
Combine with: For energy metabolism, pair it with other active B vitamins such as methylcobalamin (B12), methylfolate, and pyridoxal 5'-phosphate (P5P), plus niacin or niacinamide and a B-complex for balanced cofactor support. Magnesium, CoQ10, and L-carnitine or alpha-lipoic acid support mitochondrial function. If the user still wants cellular and DNA repair support, consider adding NAD+ precursors (NMN or nicotinamide riboside), with TMG (betaine) for methylation balance, and antioxidants such as N-acetylcysteine or glutathione, which depends on riboflavin-dependent glutathione reductase. Zinc and omega-3s are other common additions. Consult a healthcare provider, especially if taking medications.
Possible side effects: No major side effects reported in the studies; riboflavin commonly turns urine bright yellow.
