At two servings a day, this product gives 600 milligrams of pantethine, a full dose compared with the studies. In a placebo-controlled trial of 120 adults at low to moderate heart risk on a healthy diet, pantethine at 600 milligrams a day for 8 weeks and then 900 milligrams lowered total cholesterol by 3 percent and bad cholesterol by 4 percent more than placebo [1]. A smaller placebo-controlled trial of 32 adults found lower total, bad and other harmful cholesterol, with bad cholesterol down about 11 percent [2]. In adults with moderately high triglycerides, 600 milligrams a day for 8 weeks cut triglycerides by 16.5 percent [3]. A survey of 1045 people with high cholesterol taking about 900 milligrams a day found improved blood fats and very good tolerability [4], and older smaller studies also found lower cholesterol [5]. The product also gives 600 milligrams of pantothenic acid as calcium pantothenate. One older study in heart patients found calcium pantothenate had favourable effects on blood fats [6], but other trials of pantothenic acid, usually within milk or multivitamins or given with choline, found no cholesterol change [7][8][9]. Animal and cell studies are never counted.

Customers reviewing 300 mg pantethine products, not specifically The Vitamin Shoppe's, mostly buy them for cholesterol management. Many are a med-intolerant or want a gentler, more natural alternative to meds. Reviewers describe the supplement as well tolerated, with no side effects, and as a convenient daily option. They often cite the clean ingredients and the trust they place in established brands. Several say it fits alongside diet and lifestyle changes to support healthy lipids. A second group takes it for energy, adrenal and stress support. These reviewers say it relieves fatigue and helps them cope with stress. One person with an autoimmune condition took 300 mg twice daily. They reported more energy and better stress resilience, and planned to buy in larger quantities. Others describe feeling active and mentally clear within hours, or feeling more even-tempered and free of daytime crashes. Some of these reviewers had not tolerated a full B-complex and turned to pantethine instead of plain B5. Value and quality come up often, with several reviewers calling the price and quality good. There is some skepticism. One reviewer hoping to come off a med said they were also considering cheaper niacin (B3), and that it might work as well. Some users note that the 300 mg dose is on the low side, so reaching the studied 600 to 900 mg daily means taking several servings and buying more bottles. Beyond that, the feedback has few complaints about taste, pill size or side effects. Because the sample was small and drawn from other brands, check Vitamin Shoppe's own reviews before buying.

Biohacker Summary

Members Only5 of 10 recommend it
5 of 10 biohackers on our panel recommend it

Only two of the ten panelists spoke about pantethine, the main ingredient, so there is little to go on. One recommends it as a form of vitamin B5, though the statement of what it helps with is unclear, and the other takes 600 mg twice a day for cholesterol. Neither mentioned a preferred form beyond pantethine itself, a pairing, or a caution about it. Pantothenic acid, a smaller ingredient in this product, drew comments from several panelists, who mostly recommend it for fat burning, adrenal support, fatigue and repairing the gut lining, while one warns that high doses can dry the skin and another says that taking it in massive single doses can create shortfalls in other nutrients such as biotin.

Body systems: Cardiovascular system, Metabolic system

Combine with: For cholesterol support, pantethine pairs well with omega-3 fish oil (EPA/DHA) for triglycerides, soluble fiber such as psyllium or beta-glucan for LDL, plant sterols, CoQ10 if on a med, and niacin or bergamot under medical guidance. For the regularity goal, use products designed for it: psyllium husk or partially hydrolyzed guar gum for long-term stool bulk and softness, magnesium citrate or oxide for gentle osmotic support, and probiotics like Bifidobacterium lactis BB-12 or B. lactis HN019. Also drink plenty of water and eat fiber-rich foods. Check with a healthcare provider before combining supplements, especially if you take lipid-lowering or other medications.

Possible side effects: A large survey reported very good tolerability for pantethine; no major side effects reported.
