What was announced

Life Biosciences announced interim data from its Phase 1 clinical trial of ER-100. The company reported that a single intravitreal administration of ER-100 along with daily oral doxycycline was well-tolerated through Day 56 in three participants with open angle glaucoma.

No dose-limiting toxicities, adverse events of special interest, or serious adverse events were reported. All reported adverse events were related to the procedure, steroid, or pre-existing conditions, not directly associated with ER-100.

Preliminary signals of improvement in visual field testing were observed in two of the three participants. In these two participants, at least 28% of points in their visual fields showed a greater than 4 dB improvement.

The Data Safety Monitoring Board recommended escalating to Dose Level 2.

How it was done

This was a first-in-human, Phase 1, open-label clinical trial evaluating the safety and tolerability of ER-100 in adults with optic neuropathies.

ER-100 uses controlled expression of three transcription factors (OCT4, SOX2, KLF4), delivered via a single intravitreal injection. OSK expression was induced by systemic doxycycline administered for 8 weeks (56 days). An oral prednisolone taper was administered to minimize the risk of inflammation.

What it does not show

These are early, interim results from a small, open-label Phase 1 clinical trial with only three participants. The study was designed to evaluate safety and tolerability, not effectiveness. The results were announced by the company and have not been independently reviewed.

What happens next

The trial is ongoing, and the Data Safety Monitoring Board recommended escalating to Dose Level 2.
